Selection von Mycelium Strains

If you cultivate mushroom mycelium on agar medium there will appeer differnt form of mycelium. The two main forms are: rhizomorph mycelium and fluffy myclelium (looks like cotton). For further cultivation and introduction of fruiting only the rhizomorph mycelium is suitable. The rhizomorph mycelium looks like the roots of plants. The primoderia which later become fruit bodies are built from it. If you cultivate mushroom cultures on petri dishes, you have to selekt the fluffy mycelium from the rhizomorph. Only the rhizomoprh growing mycelium is used for further cultivation.

General Procedure:

As soon as your petri dishes are compeltely colonized, you can select it. Do not wait to long, otherwise the fluffy mycelium may grow over the rhizomorph strains and that means, that the petri dish is unuseable.

This petri dish is ready for selection.

As in every part of mushroom cultivation it is relevant for the success that you work clean, sterile and fast. Spring clean and desinfect your hands and the workspace bevor starting the work. We recommend the usage of latex gloves, hairnet and facemask as the working in front of a HEPA filter or inside a glovebox.

If you want to select more different petri disches at once, it is very important that you clean and desinfect your hands and workspace every time bevore you take a new mycelium culture. Please also change your latex gloves and the scalpell blade. To get the scalpell clean hold it into the flame of an alcohol lamp till the forefornt glows. Let it cool down to room temperature then insert an new (sterile) blade.

CAUTION: LATEX GLOVES AND DESINFECT ARE HIGH INFLAMMABLE. BURN HAZARD

This petri dish is selected for the first time,

and this one is selcted for the second time.

Now take the mycelium and find a rhizomorph section. Cut with the scalpell a few pieces of 3 x 3 mm from the mycelium and place each one on a petri disch filled with agar medium. Seal the ready inoculated petri dishes immediately with parafilm. Please be carefully when you move them to avoid that the mycelium piece slips arround!

TIP: Hold the petri disches which you want to select in front of a lamp. So you will find the rhizomorp strains easier!

We recommend to make a few petris from each mycelium. Even under controlled laboratory athomsphere a failure of 10 % will be normal. Don´t dismiss if you have failures of 25 % or more when you start to work with clones. For beginners this is normal manly if you don´t work in a sterile laboratory. Please note that it can also happen that the selected mycelium mutates and starts to grow fluffy. In this case the petri disch gets unusable.

The mycelium have to be selected as long as it grows uniformly rhizomorph.

Usually the petri disches are marked with date, species and strain, maybe add a clone-number. (waterproof pens, "edding pens" or CD-marker are suitable to write on a petri dish.) To start the mycelium growing, store the petri disches at the spwan-run temperatur. Mostly it will be necessary to select the mycelium for mor times bevor you get unifromly rhizomorph growing petri dishes.

As soon as the mycelium grows unifromly rhizomorph, it can be used for inoculation of grain.
