Production of spore syringes



Mainly for hobby mycologists the cultivation with spore syringes is well proven. Smal quantities of substrate or grain (mostly rye, colonized by a fungus wich is used for inoculaton of a fruiting-substrate) can be easily inoculatet with spore syringes. Even if you could not work ins perfect sterile athomosphere the failures by contamination will be very low.

General Procedure:

We recommend to use 100 ml water and 1 sporeprint per each glass. Fill the glasses with water and and cover the top with aluminium foil sterilize them at 121 °C / 250 °F / 15 psi / 1,05 bar for 30 minutes. Time runs from the moment pressure has reached reqired level. To make pressure exchange between the glasses and the pot possible, please make a hole (1 cm) in the cover of the glasses. To minimize contamination risk, you can use a filter disk for the glasses (below the cover). We recommend to work in front of a Hepafilter or inside a Glovebox / Inoculationbox.



To keep contamination risk low, please work clean! Use a new, sterile skalpell blade for each spore print.



As soon as the glasses with water have cooled down to room temperature they could be processed. Take a spore print and scrape off the spores into the galss. Please use a scalpell bis sterile blade. It is important hat you donīt touch the spore print. Work clean and fast. Spring clean your hands, wear latex gloves, hairnet and facemask and desinfect the workspace and your hands.

After the spores were scraped into the glass, close it with an intact cover (without a hole) and shake it well to dispense the spores equally in the water. To soak the spore soulution into the syringes put again the cover with the hole on the glass. We recommend to fill 10 ml into each syringe. Donīt touch the needle! If the needle get in contact with any contamination, please heat it on a alcohol lamp or similar till it glowes, let it cool down and cover it fast.





We recommend to store the syringes in an airproof bag (zipp-lock bag). Stored in a cool (max. 20 °C) and dark place spore syringes are about 1 year germinable.

For testing the germination capacity, put a few mililiter (1 or 2 drops) of the spore solution on a agar filled petri dish. Depending on the strain there should appear mycelium growing after a few days up to 1 week.
